The symptoms suggest that one or more fields have simple validation (i.e.
require a validation) set on them, not necessarily the Subject field. Did
you perhaps add a field, set validation, then later remove that field?
It's best never to use simple validation, but instead to use a validation
formula and an explicit validation message to the user that gives the name
of the field under validation.

I have a custom form region in Outlook 2007 defined. It worked fine as
long as I only used Custom Fields bound to my controls. Now I decided to
use the "Subject" Field on the form. Since then. When I create a new item
and choose "Save and Close" I get an error message that "one field on the
form requires a value" although I have entered a value into the Subject
Control. I really have no idea what the problem is.
